Roivant and Priovant reported that brepocitinib met the primary endpoint in both CLARITY-1 and CLARITY-2, their Phase 3 studies in non-infectious uveitis. Median time to treatment failure exceeded 48 weeks in the treatment groups, compared with approximately 12 weeks on placebo.
Across the pooled studies, 62% of patients receiving brepocitinib achieved steroid-free quiescence at week 48, against 20% receiving placebo. The companies also reported benefits on key secondary endpoints and no new safety signals.
Priovant intends to submit a supplemental US application in the first half of 2027. The results therefore establish a regulatory opportunity rather than an approval for this indication.
The commercial appeal is sustained control of inflammation while reducing reliance on steroids. Two successful pivotal trials provide a stronger foundation than a single exploratory readout. Nevertheless, the company's suggestion of best-in-class potential goes beyond what placebo-controlled results alone can prove. Regulators and clinicians will assess the complete efficacy and safety package before determining its place in treatment.
